Bacopin® obtained from the leaves of Bacopa monniera (Bacopa monnieri) is commonly known as a natural memory enhancing herb.

Traditional Knowledge
B. monniera, known colloquially as ‘Brahmi’ has been used traditionally as a brain tonic to enhance memory development, learning, concentration and to alleviate symptoms associated with anxiety and epileptic disorders. The name ‘Brahmi’ is derived from the word ‘Brahma’ which translates to ‘mythical creator’ in Hindu mythology. According to Indian philosophy, the brain is considered to be the center of creative activity, thereby rightly naming a nutrient that improves this ability as ‘Brahm’.
Mechanism of Action
Repairing damaged neurons: Bacosides are reported to promote nerve impulse transmission by repairing damaged neurons. This is done by enhancing kinase activity, neuronal synthesis and restoration of synaptic activity. (Singh HK et. al (1997). Indian J. Pharmacol., 29:5359-365)
Inhibiting Acetylcholinesterase (AChE): Acetylcholinesterase hydrolyzes the neurotansmitter Acetycholine. It is found at neuromuscular junctions and cholinergic brain synapses, where its activity serves to terminate synaptic transmission. Higher levels of AChE have been reported to play a major role in the progression of Alzheimer’s disease.
B. monniera suppresses the function of AChE in the cerebral cortex, particularly in the parietal cortex and hippocampus, leading to increased availability of Acetylcholine. This facilitates enhanced attention, memory and cognitive processing thereby resulting in enhanced working memory.
Preclinical study
The oral dose (100 mg/kg body weight) of B. monniera extract inhibited acetylcholinesterase differentially in various parts of the animal brain viz. Cerebral cortex (51.6%) > Cerebellum (51%) > Pons (44%) > Thalamus (41.6%) > Hippocampus (38.1%) > Brain stem (34.3%) > Striatum (24.9%).

Clinical studies
In a randomized double-blind placebo-controlled study, 60 healthy elderly volunteers (mean age 62) consisting of 23 males and 37 females, received either a standardized extract of B. monniera (300 and 600 mg) or placebo daily for 12 weeks.
B. monniera at a dose of 300mg/day was found to

B. monniera extract (300mg/kg) suppresses AChE activity resulting in enhanced cholinergic function, which in turn enhances attention and memory processing and gives rise to increased working memory.

These results suggest that B. monniera can improve attention, cognitive processing, and working memory partly via the suppression of AChE activity.
